Computational identification of potential bioactive compounds from Triphala against alcoholic liver injury by targeting alcohol dehydrogenase.

Alcoholic liver injury resulting from excessive alcohol consumption is a significant social concern. Alcohol dehydrogenase (ADH) plays a critical role in the conversion of alcohol to acetaldehyde, leading to tissue damage. The management of alcoholic liver injury encompasses nutritional support and, in severe cases liver transplantation, but potential adverse effects exist, and effective medications are currently unavailable. Natural products with their potential benefits and historical use in traditional medicine emerge as promising alternatives. Triphala, a traditional polyherbal formula demonstrates beneficial effects in addressing diverse health concerns, with a notable impact on treating alcoholic liver damage through enhanced liver metabolism. The present study aims to identify potential active phytocompounds in Triphala targeting ADH to prevent alcoholic liver injury. Screening 119 phytocompounds from the Triphala formulation revealed 62 of them showing binding affinity to the active site of the ADH1B protein. Promising lipid-like molecule from Terminalia bellirica, (4aS, 6aR, 6aR, 6bR, 7R, 8aR, 9R, 10R, 11R, 12aR, 14bS)-7, 10, 11-trihydroxy-9-(hydroxymethyl)-2, 2, 6a, 6b, 9, 12a-hexamethyl-1, 3, 4, 5, 6, 6a, 7, 8, 8a, 10, 11, 12, 13, 14b-tetradecahydropicene-4a-carboxylic acid showed high binding efficiency to a competitive ADH inhibitor, 4-Methylpyrazole. Pharmacokinetic analysis further confirmed the drug-likeness and non-hepatotoxicity of the top-ranked compound. Molecular dynamics simulation and MM-PBSA studies revealed the stability of the docked complexes with minimal fluctuation and consistency of the hydrogen bonds throughout the simulation. Together, computational investigations suggest that (4aS, 6aR, 6aR, 6bR, 7R, 8aR, 9R, 10R, 11R, 12aR, 14bS)-7, 10, 11-trihydroxy-9-(hydroxymethyl)-2, 2, 6a, 6b, 9, 12a-hexamethyl-1, 3, 4, 5, 6, 6a, 7, 8, 8a, 10, 11, 12, 13, 14b-tetradecahydropicene-4a-carboxylic acid from the Triphala formulation holds promise as an ADH inhibitor, suggesting an alternative therapy for alcoholic liver injury.

LC-ESI-QTOF-MS/MS Identification and Characterization of Phenolic Compounds from Leaves of Australian Myrtles and Their Antioxidant Activities

Phenolic compounds, present in plants, provide substantial health advantages, such as antioxidant and anti-inflammatory properties, which enhance cardiovascular and cognitive well-being. Australia is enriched with a wide range of plants with phytopharmacological potential, which needs to be fully elucidated. In this context, we analyzed leaves of aniseed myrtle (Syzygium anisatum), lemon myrtle (Backhousia citriodora), and cinnamon myrtle (Backhousia myrtifolia) for their complex phytochemical profile and antioxidant potential. LC-ESI-QTOF-MS/MS was applied for screening and characterizing these Australian myrtles’ phenolic compounds and the structure–function relation of phenolic compounds. This study identified 145 and quantified/semi-quantified 27 phenolic compounds in these Australian myrtles. Furthermore, phenolic contents (total phenolic content (TPC), total condensed tannins (TCT), and total flavonoids (TFC)) and antioxidant potential of phenolic extracts from the leaves of Australian myrtles were quantified. Aniseed myrtle was quantified with the highest TPC (52.49 ± 3.55 mg GAE/g) and total antioxidant potential than other selected myrtles. Catechin, epicatechin, isovitexin, cinnamic acid, and quercetin were quantified as Australian myrtles’ most abundant phenolic compounds. Moreover, chemometric analysis further validated the results. This study provides a new insight into the novel potent bioactive phenolic compounds from Australian myrtles that could be potentially useful for functional, nutraceutical, and therapeutic applications.

Sources of Carotenoids in Amazonian Fruits

Epidemiological studies have shown that a diet rich in bioactive components significantly reduces cardiovascular disease incidence and mortality. In this sense, there is a need for meta-analytical research that confirms this phenomenon and increases specific knowledge about certain bioactive compounds such as carotenoids. Thus, this systematic review and meta-analysis aim to disseminate knowledge about the sources of carotenoids in fruit consumed in the north of Brazil which are outside the Brazilian trade balance. A systematic review and a meta-analysis following the PRISMA guidelines were conducted based on a random effects synthesis of multivariable-adjusted relative risks (RRs). Searches of seven sources were carried out, including PubMed, Science Direct from Elsevier, Web of Science, Scielo, Eric Research and Google Scholar databases. The systematic review was guided by a systematic review protocol based on the POT strategy (population, outcome and type of study) adapted for use in this research. Mendeley was a resource used to organize and manage references and exclude duplicates of studies selected for review. In this review, we present the potential bioactive compounds concentrated in little-known fruit species from the Amazon and their benefits. Consuming fruits that are rich in notable constituents such as carotenoids is important for the prevention of chronic non-communicable diseases through anti-inflammatory and anticoagulant properties, as well as antivirals, immunomodulators and antioxidants agents that directly affect the immune response.

Centipeda minima (L.) A. Braun & Asch. and its representative active compound alleviate DSS-induced ulcerative colitis via inhibition of NLRP3 inflammasome activation and regulation of gut microbiota

Ulcerative colitis (UC) is a prevalent chronic inflammatory bowel disease (IBD), which is related to the occurrence of inflammation and intestinal flora disorder. Centipeda minima (L.) A. Braun & Asch. (CM for short hereafter) is commonly used to treat IBD in folk medicine. However, the anti-UC mechanisms of CM and its representative active compound(s) are not yet fully understood. Here, we sought to investigate the mode of action and the underlying mechanism of CM as an anti-UC agent, as well as to identify its potential bioactive compound(s). Results showed that 50% ethanol extract of CM (CME) significantly improved the pathological morphology and reduced the protein levels of NLRP3 both in DSS-induced UC mice model and lipopolysaccharide (LPS)-stimulated RAW264.7 cell model. Additionally, it decreased the release of proinflammatory cytokines including TNF-α, IL-18 and IL-1β. These results suggest that the anti-UC effect of CM is at least partially attributed to the inhibition of NLRP3 inflammasome activation. Furthermore, luteolin, a representative active compound in CM, exhibited significant improvement of the pathological changes, reduced the release the of proinflammatory cytokines, downregulated NLRP3 inflammasome activation, enhanced intestinal barrier, and modulated gut microbiota in vivo. This study provides a molecular rationale for the traditional use of CM in treating UC, and offers a pharmacological basis for the development of modern anti-UC agents derived from CM.

Nutritional profiling and in silico analysis of pharmacological activities from local rice Pulu Mandoti fermented with Pleurotus spp.

Pulu Mandoti, a local red rice (Oryza sativa L.) variety popular among Sulawesi residents, has gained recognition for its perceived health benefits, especially as a preferred dietary option for individuals with diabetes or those seeking to prevent obesity. Given the increasing consumption of mushrooms, particularly Pleurotus species, renowned for their nutritional and medicinal attributes, this study delves into the transformative effects of Pleurotus spp. fermentation on Pulu Mandoti, the indigenous rice variety. Proximate analysis disclosed elevated dry matter (91.99 ± 0.61%), crude protein (8.55 ± 0.15%), and crude fat (1.34 ± 0.05%) in Pleurotus cystidiosus fermentation compared to Pleurotus ostreatus and Pleurotus djamor. Concurrently, antioxidant and antidiabetic activities were notably improved in all Pleurotus fermentations. Pulu Mandoti fermented with P. cystidiosus outperformed other treatments, aligning with molecular docking results pinpointing 11-Eicosenoic acid, methyl ester, and butylated hydroxytoluene as optimal interactors with antioxidant receptors 5O0x and 2CKJ. Butylated hydroxytoluene demonstrated interactions with the antidiabetic receptor 2QV4, along with 9-Octadecenoic acid, methyl ester. These compounds, previously unreported in Pleurotus, displayed promising attributes as antioxidants and antidiabetic agents. Furthermore, the investigation delved into the fatty acid profiles, emphasizing the diverse range of potential bioactive compounds in fermented Pulu Mandoti. The findings of this research present a potential functional food rich in natural antioxidants and antidiabetic compounds, highlighting the yet undiscovered capabilities of Pleurotus spp. fermentation in augmenting the nutritional composition and bioactivity of indigenous rice varieties, specifically Pulu Mandoti.

Antioxidant Effects and Potential Molecular Mechanism of Action of Diplocyclos palmatus (L.) C. Jeffrey Fruits Based on Systematic Network Pharmacology with Experimental Validation

Oxidative stress arises from an imbalance between the production of reactive oxygen species (ROS) and the ability of biological systems to detoxify these reactive intermediates or repair the resulting damage. Diplocyclos palmatus (D. palmatus) is a medicinal plant, whose fruits has been reported to have antioxidant activity due to its potent bioactive compounds, and holds promise for mitigating oxidative stress and its associated diseases. Therefore, this study aims to elucidate the bioactive compounds present in D. palmatus via Gas Chromatography-Mass Spectrometry (GC-MS) analysis and evaluate their antioxidant activity via different in-vitro antioxidant assays such as, DPPH free radical scavenging assay, ABTS radical scavenging assay, ferric reducing antioxidant power assay, hydroxyl free radical scavenging assay and reactive nitrogen oxide scavenging assay. Furthermore, systematic network pharmacology, molecular docking and molecular dynamics were utilized to identify major bioactive compounds and their interactions with key proteins implicated in oxidative stress to understand the molecular mechanism of action of D. palmatus fruits. The obtained results indicated that the fruit extract had highest antioxidant activity against hydroxyl free radicals (IC50 = 476.76 µg/mL) followed by nitric oxide radicals (IC50 = 509.06 µg/mL), DPPH• (IC50 = 601.88 µg/mL), ABTS•+ (IC50 = 709.37 µg/mL) and FRAP (IC50 = 1141.76 µg/mL). In network pharmacology analysis, bioactive compounds of D. palmatus fruits were found to target numerous major cellular proteins (AKT1, GAPDH, ALB, CASP3, PTGS2, PPARG, STAT3, EGFR, ESR1 and MMP9), with a notable impact on the estrogen signaling pathway. Molecular docking and dynamics analysis further validated the antioxidant potential of D. palmatus fruits. Overall, this integrative study sheds light on the antioxidant potential of D. palmatus fruits and elucidates the molecular mechanisms underpinning their antioxidative actions. These findings not only deepen the understanding of D. palmatus phytochemistry but also highlight its therapeutic potential as a natural antioxidant agent for combating oxidative stress-related disorders.

Polyphyllin I Sensitizes Cisplatin-Resistant Human Cervical Cancer Cells to Cisplatin Treatment

Cervical cancer (CC) is a common gynecological malignancy, and improving cisplatin sensitivity has become a hot topic in CC chemotherapy research. Polyphyllin I (PPI), a potent bioactive compound found in Rhizoma Paridis, known for its anticancer properties, remains underexplored in CC resistance. In this study, we evaluated PPI’s impact on cisplatin-resistant CC cells and elucidated its underlying mechanism. Our findings reveal that PPI enhances the sensitivity of cisplatin-resistant CC cells to the drug, promotes apoptosis, and inhibits cell migration. Mechanistically, PPI was found to regulate p53 expression and its target genes, and suppressing p53 expression reverses PPI’s sensitizing effect in drug-resistant CC cells. In conclusion, PPI showed promise in sensitizing cisplatin-resistant human CC cells to cisplatin treatment, suggesting that it could serve as a potent adjunct therapy for cervical cancer, particularly for cases that have developed resistance to cisplatin, thereby providing a promising basis for further clinical investigation into PPI for enhancing the efficacy of existing chemotherapy regimens in resistant cervical cancer.

The Fucaceae family of marine brown algae includes Ascophyllum nodosum. Fucosterol (FSL) is a unique bioactive component that was identified through GC-MS analysis of the hydroalcoholic extract of A. nodosum. Fucosterol's mechanism of action towards hepatocellular cancer was clarified using network pharmacology and docking study techniques. The probable target gene of FSL has been predicted using the TargetNet and SwissTargetPred databases. GeneCards and the DisGNet database were used to check the targeted genes of FSL. By using the web programme Venny 2.1, the overlaps of FSL and HCC disease demonstrated that 18 genes (1.3%) were obtained as targeted genes Via the STRING database, a protein-protein interaction (PPI) network with 18 common target genes was constructed. With the aid of CytoNCA, hub genes were screened using the Cytoscape software, and the targets' hub genes were exported into the ShinyGo online tool for study of KEGG and gene ontology enrichment. Using the software AutoDock, a hub gene molecular docking study was performed. Ten genes, including AR, CYP19A1, ESR1, ESR2, TNF, PPARA, PPARG, HMGCR, SRC, and IGF1R, were obtained. The 10 targeted hubs docked with FSL successfully. The active components FSL of ASD, the FSL, are engaged in fatty liver disease, cancer pathways, and other signalling pathways, which could prove beneficial for the management of HCC.

Cytotoxic Potential of Berunok Sea Cucumber (Paracaudina australis) Against Breast Cancer Cells (T47D)

Background Sea cucumbers can be explored as alternative raw materials by the pharmaceutical and fishery industries as anticancer agents because they contain potential bioactive compounds. Methods This study aimed to determine the anticancer activity of Paracaudina australis extract against breast cancer cells (T47D) using an MTT assay. The secondary metabolites found in P.australis are steroids, terpenoids, saponins, and phenolics. The Thin-Layer Chromatography test results are indicated by Rf values, and steroid compounds in the ethyl acetate fraction are included in the standard Rf values. The isolates obtained were identified by Gas Chromatography-Mass Spectrometry, Fourier-Transform Infrared Spectroscopy, High-performance liquid chromatography, and UV-Visual spectrophotometry. Results The isolated compounds were Dihydrocholesterol and Cholestan-3-ol, with a molecular formula (C27H48O). The ion weight and molecular mass of the compound were m/z 388,7. Conclusions This compound may be responsible for the anticancer activity of P.australis. The IC50 of Isolate F4 was 25,3 μg/ml, and IC50 of Isolate F7,8 was 13,76 μg/ml.

Nutritional evaluation and GC–MS analysis of bioactive compounds present in methanol extracts of dried fruit pericarp of garcinia pedunculata Roxb

This study was conducted to assess the nutritional and phytochemical properties of methanol extracts derived from the dried fruit pericarp of Garcinia pedunculata. Samples of the plant were obtained from the Bodoland Territorial Region (BTR), Assam, India due to the lack of study findings on Garcinia pedunculata from this region in the existing literature. Nutritional evaluation has revealed that Garcinia pedunculata is a notable source of various nutrients  including fibre (15.67 ± 0.05 g/100g), protein (3.97 ± 0.06 g/100g), carbohydrates (80.10 ± 2.48 g/100g)  and ascorbic acid (98.16 ± 4.46 mg/100g). Furthermore, the results of the gas chromatography coupled with mass spectrometry analysis (GC-MS) of the methanol extract revealed a diverse range of chemical compounds with both high and low molecular weight. GC-MS analysis showed bioactive compounds like cyclopentanecarboxylicacid, 2-oxo-, ethyl ester with a peak area of 23.60%followed by 1,3- dioxepane, 2-pentadecyl- with a peak area of 22.39%followed by N-[Carboxymethyl ]maleamic acid with a  peak area of 10.37%and  butanedioic acid,3-hydroxy-2,2-dimethyl, dimethyl ester, (R) with a peak area of 10.91%. The extracts exhibit significant amount of various naturally occurring bioactive compounds. Therefore, the existence of potential bioactive compounds and nutritional constituents can be suitably exploited for future studies.

The effect of butterfly pea (Clitoria ternatea L.) and roselle (Hibiscus sabdariffa L.) powder addition on the physicochemical and sensory properties of white chocolate ganache

Ganache is a mixture of chocolate and cream commonly utilized as fillings or glazing. Butterfly pea and roselle are edible flowers with potential bioactive compounds that could improve ganache functionality. This research aimed to understand the effect of butterfly pea and roselle powder addition with various concentrations (1%, 1.5%, and 2% (w/v)) on white chocolate ganache’s physical and sensory properties. The samples were analyzed for their water content, color profile, water activity, total soluble solids, and hedonic scores. Results showed that the butterfly pea-added ganache had a higher water content and activity than the roselle-added ganache. However, the total soluble solid analysis showed a reverse trend. Addition of 2% edible flower powder decreased the moisture content of ganache up to 2%. Increasing the concentration of butterfly pea powder resulted in higher blueness intensity, while in roselle ganache, there was no significant effect on the redness. The hedonic test showed that the difference in edible flower types and concentration resulted in no significant difference in panelists’ acceptance of all tested sensory attributes.

In silico molecular docking study and in vitro evaluation of antioxidant activity in Kacip Fatimah

In modern drug and supplement design, molecular docking is employed to comprehend molecular interaction. Nevertheless, there is currently scarce literature available on Kacip Fatimah regarding the molecular docking analysis of its secondary metabolites for antioxidant activity. To identify molecules that effectively bind to antioxidant enzymes, docking with scoring function can be employed. Consequently, this study seeks to identify potential bioactive compound(s) responsible for antioxidant properties through in silico molecular docking and validated through in vitro antioxidant activity. To fulfil the objective, the study involved characterizing the binding interactions between the ligands of secondary metabolites in Kacip Fatimah and enzymatic antioxidants. Using catalase (CAT) as the target receptor and secondary metabolites as ligands, the interaction aimed to identify the optimal binding scoring of secondary metabolites. The findings indicate that myricetin, a flavonoid, displayed the most significant binding energy during molecular docking with 2CAG catalase, establishing it as the most potent antioxidant compound in Kacip Fatimah. Among all the docked secondary metabolites, flavonoids demonstrated superior antioxidant activity, as they produced the lowest energy interaction in the scoring function. The in silico findings were further substantiated by in vitro antioxidant assessments, confirming significant antioxidant activity through DPPH scavenging and protective effects on HepG2 cells. In summary, the in silico and in vitro study indicated that Kacip Fatimah’s secondary metabolites show potential as effective antioxidant agents.

Effects of Elicitation on Invitro Regeneration of two Tomato (Solanum lycopersicum L.) Cultivars in Tissue Culture

Exploring alternative avenues, in vitro culture emerges as a promising option for potential bioactive compound sources. However, compared to intact plants, only a few cultures demonstrate efficient synthesis of secondary metabolites. Elicitors have gained prominence as stress agents for enhancing in vitro micropropagation in specific tissues, organs, and cells. Recent advancements in plant tissue culture involve elicitors, opening new possibilities for in vitro production of crucial food crops. This research aimed to investigate the impact of three elicitors (Activane®, Micobiol®, and Stemicol®) on germination and in vitro multiplication of two tomato cultivars explants, employing both direct and indirect in vitro organogenesis. Among the texted elicitors, Micobiol® emerged as a successful elicitor, promoting optimal seed germination, survival, and 100% growth compared to the 80% in the control group. Further, Activane® exhibited a favourable induction response and achieved 96%, 95%, and 100% in weight and diameter of callus, yet various elicitor concentrations did not exert significant influence across treatments. In conclusion, an effective disinfection and in vitro implantation of tomato seeds ensured successful germination, promoting seedling survival and growth. Various elicitors positively impacted in vitro organogenesis, particularly in root induction, with higher survival percentages in acclimatized plants. The study guides future research on elicitor treatments for large-scale tomato in vitro propagation, emphasizing the need to identify optimal elicitor concentrations.

The use of ionic organic solids, such as 1,3-bis(carboxymethyl)imidazolium chloride as a catalyst has allowed the Michael addition of N-heterocycles to chalcones. This methodology has been applied to the unique preparation of the potential bioactive compound 1-(3,4-dimethoxyphenyl)-3-(4-methoxyphenyl)-3-(1H-1,2,4-triazol-1-yl)propan-1-one with moderate yield, due to the retro-Michael reaction. Both synthetic reactions (i.e., preparation of chalcone and triazole Michael-addition to chalcone) have good green metrics.

Identification of anti-fibrotic and pro-apoptotic bioactive compounds from Ganoderma formosanum and their possible mechanisms in modulating TGF-β1-induced lung fibrosis

Ethnopharmacological relevanceThe Compendium of Materia Medica and the Classic of Materia Medica, the two most prominent records of traditional Chinese medicine, documented the therapeutic benefits of Ganoderma sinense particularly in addressing pulmonary-related ailments. Ganoderma formosanum, an indigenous subspecies of G. sinense from Taiwan, has demonstrated the same therapeutic properties. Aim of the studyThe aim of this study is to identify bioactive compounds and evaluate the potential of G. formosanum extracts as a novel treatment to alleviate pulmonary fibrosis (PF). Using an in-house drug screening platform, two-stage screening was performed to determine their anti-fibrotic efficacy. Methods and materialsG. formosanum was fractionated into four partitions by solvents of different polarities. To determine their antifibrotic and pro-apoptotic properties, the fractions were analyzed using two TGF-β1-induced pulmonary fibrosis cell models (NIH-3T3) and human pulmonary fibroblast cell lines, immunoblot, qRT-PCR, and annexin V assays. Subsequently, transcriptomic analysis was conducted to validate the findings and explore possible molecular pathways. The identification of potential bioactive compounds was achieved through UHPLC-MS/MS analysis, while molecular interaction study was investigated by multiple ligands docking and molecular dynamic simulations. ResultsThe ethyl acetate fraction (EAF) extracted from G. formosanum demonstrated substantial anti-fibrotic and pro-apoptotic effects on TGF-β1-induced fibrotic models. Moreover, the EAF exhibited no discernible cytotoxicity. Untargeted UHPLC-MS/MS analysis identified potential bioactive compounds in EAF, including stearic acid, palmitic acid, and pentadecanoic acid. Multiple ligands docking and molecular dynamic simulations further confirmed that those bioactive compounds possess the ability to inhibit TGF-β receptor 1. ConclusionPotential bioactive compounds in G. formosanum were successfully extracted and identified in the EAF, whose anti-fibrotic and pro-apoptotic properties could potentially modulate pulmonary fibrosis. This finding not only highlights the EAF's potential as a promising therapeutic candidate to treat pulmonary fibrosis, but it also elucidates how Ganoderma confers pulmonary health benefits as described in the ancient texts.

Potential bioactive compound and hub gene identification of endometrial carcinoma using systems biology

Endometrial carcinoma is a frequent cancer of the female genital tract. Endometrial carcinoma accounts for 97% of all uterine malignancies and 3 % of sarcomas that develop from the endometrium’s glands. Endometrial cancer is frequently found in its early stages since most women quickly report postmenopausal vaginal hemorrhage. The need for more advanced medications to improve survival in such situations is still unfulfilled. As a result, there is growing interest in employing an herbal treatment to treat endometriosis, which seems to be an effective strategy. We have discovered a few unintended targets (ligands) in our investigation that are active components of common therapeutic herbs. The differentially expressed genes (DEG - target protein) for endometrial cancer were found using the NCBI and CIViC databases. In our investigation, the protein used for docking and simulation was PDB ID: 3THW. Using the Cytoscape server, the gene-encoding protein network has been identified. It was discovered that the Protein 3THW's binding energy to the bioactive substance (Asarone) was −7.15 Kcal/mol. It was discovered that the crucial interacting amino acid residues were ILE648, PHE650, ILE651, VAL802, TYR815, VAL817. The properties of the pharmaceutical target are further investigated by employing a molecular simulation study for 100 ns with NAMD software. Low RMSD and SASA (Solvent accessible surface area), high RMSF, High hydrogen bonds, between Asarone and MSH2 demonstrated their potency as endometrial cancer inhibitor compounds. Based on these analyses we infer that the bioactive substances originating from medicinal plants may be an effective treatment for endometrial cancer. Communicated by Ramaswamy H. Sarma

Crystal structure and properties of new benzimidazopyrido[2,3-b]indole-6-carbonitriles

The structure of representatives of a new heterocyclic system, 12H-benzimidazo[1',2':1,6]pyrido[2,3-b]indole-6-carbonitriles, potential bioactive compounds, is elucidated by X-ray diffraction. The compounds exhibit luminescence in the blue region of the spectrum with quantum yields close to unity, which makes them promising phosphors for application in modern emitting devices.
